Johnson & Johnson said Oct. 25 that 12 of 16 patients with measurable advanced prostate cancer in its 50-mg JNJ-6143 Phase 1 cohort had confirmed tumor responses, a 75% rate.
An earlier EconoTimes report on imaging results gave a tally of 11 responses among 16 patients (69%). The company’s latest announcement gives 12 of 16 and does not explain how the two reports relate.
JNJ-6143, previously called HLD-0915, is an oral small molecule in Johnson & Johnson’s Regulated Induced Proximity Targeting Chimera (RIPTAC) class. Johnson & Johnson describes JNJ-6143 as a RIPTAC molecule intended to link BRD4 with the androgen receptor, a protein highly expressed in prostate cancer cells, and interfere with tumor-cell activity.
The Phase 1 study enrolled patients with metastatic disease that had progressed after at least one androgen receptor pathway inhibitor. Patients in the 50-mg group had received a median of four prior therapies.
Johnson & Johnson selected the 50-mg once-daily dose for further study. The Phase 2 trial began Sept. 16 and is recruiting.
